1. Technical requirements for the use of ordinary high pressure sterilizers (1) Before using the sterilizer, add an appropriate amount of water up to the designated line. It is recommended to use hot water that has been boiled to prevent scale buildup and reduce sterilization time. (2) Place the items to be sterilized inside the pot, ensuring they are not too tightly packed to allow proper steam circulation and effective sterilization. Avoid placing items near the sides of the pot to prevent condensation from dripping onto them. (3) Open the exhaust valve and heat the device until steam begins to escape. Close the exhaust valve after 3–5 minutes. This ensures all cold air is removed. As steam builds up, the temperature will rise with increasing pressure. Once the desired pressure is reached (typically 1 kg/cm² or 1.034×10^5 Pa), maintain it for about 20 minutes. (4) After sterilization, turn off the heat source and wait until the pressure drops to zero before opening the lid. Do not leave the items in the sterilizer for long, as condensation may form on the lid and walls, leading to moisture and potential contamination. (5) To check if the sterilization was successful, take a small portion of the medium and incubate it at 37°C for 24 hours. If no growth occurs, it is considered sterile. Similarly, slant culture media should be cooled and incubated for the same period before storage. 2. Technical requirements for the use of fully automatic high-pressure steam sterilizers (1) Check the water level in the tank and ensure it is at the maximum level indicated. (2) Confirm the water level at the bottom of the sterilizer is just below the base. Turn on the power, and the display should start flashing. Ensure the steam discharge valve is closed. (3) Place the items into the sterilizer, push the rack to the left, and make sure all beams are fully inserted into the column. Turn the handle clockwise to secure the lid properly. (4) Choose the sterilization program—either manual or automatic. Use the control panel to set the required temperature and time. (5) Press the start button. The display will flash until the internal temperature reaches 80°C, after which it will show the actual temperature. Once above 100°C, the pressure gauge will begin to indicate the internal pressure. The system will automatically maintain the set conditions for the required duration. (6) After sterilization, quickly dry the items by opening the safety valve or turning the exhaust valve to the "open" position. This helps remove any remaining moisture. However, never use this method when sterilizing liquids. (7) Once the sterilization cycle is complete and the chamber is depressurized, turn the handle counterclockwise to open the door and retrieve the sterilized items for storage.
